It proposes a two-path model: folate receptor alpha autoantibodies (FRAA) causing cerebral folate deficiency (affecting 50-70% of autistic children), and oxidative stress/neuroinflammation from environmental or genetic factors. These pathways converge during vulnerable prenatal and early childhood periods (ages 1-3), disrupting synaptic pruning and brain development.

The work spans three parts: cellular mechanisms and critical periods; genetic/environmental contributors including maternal immune activation and pollutants; and clinical applications. Key findings include that leucovorin (natural reduced folate) can bypass blocked receptors to reduce ASD risk, unlike synthetic folic acid. The book culminates in a proposal for non-invasive retinal imaging combined with biochemical screening to detect risk before symptoms emerge, enabling targeting vitamin supplementation in prenatal and infant interventions.
